The UMR GABI is made up of eight research teams and one core facilities. The scientific objectives of the UMR GABI are aimed at gaining knowledge on the genetic variability of biological functions and developing methods and tools for genetic improvement and conservation of animal populations. The UMR GABI develops research programs for farming and biomedicine, notably through the INRA-CEA LREG project that is focused on the study of the genetic sensitivity to the development of cutaneous melanoma in the porcine MeLiM model, and the mechanisms for spontaneous regression of tumors.
Research at the GIS Team is focused on the study of the genetic architecture of traits associated with health and adaptation in pigs, chickens and rabbits. Their projects use animal experimentation and genomic and integrative biology approaches.

The LREG is a group belonging to the GIS Team, comprised of three scientists and four technicians. During 2015, they hosted a doctoral student (Michaël Baco) and a postdoctoral scholar (Fany Blanc).​

RESEARCH PROJECTS
The LREG maintains a cutaneous melanoma porcine model (MeLiM model) in which tumors appear in utero or rapidly after birth, causing metastases that regress spontaneously during the first year of life of the piglets. This regression is accompanied by a loss of pigmentation of the hair coat and vitiligo. The group develops studies aimed at the genetic susceptibility of the development of cutaneous melanoma in this porcine experimental model along with mechanisms inducing a spontaneous tumor regression.
Genome Wide Association Studies (GWAS) and functional studies of gene candidates are carried forth on MeLiM porcine families in order to identify what genes are implicated in the oncogenesis of porcine melanoma.

​In parallel, comparative genetic studies are performed using the results obtained by partners from human families developing melanomas (in partnership with F. Demenais’ team, INSERM, Paris) and in dog breeds that are carriers for cutaneous melanoma (in partnership with C. André’s team, CNRS, Rennes). In order to understand the cellular and molecular mechanisms implicated in the induction of tumor regression, studies are performed on the tumor (transcriptomics analysis: RNA and microRNA) and on the antitumoral immune response of the host (phentoyping of immune cells, search for antibodies and ​functional tests).

All the studies are led in partnership with other scientists from the UMR GABI, the INRA Jouy en Josas research center (Virology and Immunology), national partners (INSERM, CNRS) or international partners (Czech Republic, Spain, Taiwan).

​EXPERIMENTAL FACILITIES

  • ​Techniques and expertise : molecular biology, genotyping, bioinformatics, cytometry, primary culture of melanocytes and tumor cells, dosage of cytokines using cytometry, immunophenotyping, histology, immunohistology, animal experimentation (pigs).
  • The team has experimental facilities at its disposition for the MeLiM pig model including three experimental pig houses and a birthing room (2 spaces), an operating room, a laboratory for sample preparation and a laboratory for autopsies.
  • ​The LREG provides scientific  (S. Vincent-Naulleau) and technical (F. Andréoletti) support to the histology laboratory of UMR GABI’s core facilities.

THE LREG coordinates or participates in projects financed by the National Cancer Institute, the Cancer league, the Research Association Against Cancer and INRA. ​
